GH_4C_1細胞で見られたオートファジ-の解析とユビキチンの生理機能について

研究概要

(1)免疫組織化学によるオートファゴゾームの生成機序の解析
オートファジ-を誘導したGH4C1細胞を、凍結超薄切片上での免疫電顕法にて解析した。カテプシンB、H、L、および1gp120に対する抗体でリソゾームを標識したところ、リソゾーム自身が細胞質、分泌顆粒、残余小体、などを取り込む像が多数見られた。このことは、この細胞に見られるオートファゴソームが、飢餓時の肝臓に見られるようなマクロオートファジ-(小胞体による包み込み)ではなく、ミクロオートファジ-(リソゾームによる食べ込み)により生成されることを示唆する。
(2)ユビキチンの局在
共焦点レーザー顕微鏡を用いた蛍光抗体法と免疫電顕法を用いてこの細胞におけるユビキチンの局在を検索したところ、核、細胞質、リソゾーム、および空胞にユビキチンの免疫反応が認められた。特に空胞内では電子密度の高い顆粒の表面に、強いユビキチンの免疫反応が認められたこと、またミクロオートファジ-によって取り込まれるオルガネラの膜表面や残余小体内の膜成分上にも認められたことより、ユビキチンがミクロオートファジ-、あるいはオートファゴゾームの成熟過程に何らかの役割を持つことが示唆される。
一方、パーコールを用いた密度勾配遠心により細胞分画を得、ウエスタンブロットによりユビキチンを解析したところ、リソゾームおよび細胞質分画に遊離型のユビキチンが認められた。また、僅かであるが膜分画(まだ特定していない)には、タンパクに結合したユビキチンと思われる高分子量のバンドも見られた。現在、細胞分画中のオルガネラを免疫電顕で解析する手法を確立している過程である。
