新規MAPKKK関連キナーゼの細胞内情報伝達における役割

研究概要	哺乳類のMAPキナーゼカスケードの上流のシグナル分子を検索する目的で、酵母のMAPKKKのアミノ酸配列を基にRT-PCRを行い、MUK1、MUK2、MUK3という3種の新規キナーゼを単離した。MUK1は、酵母の胞子形成に関与するMAPキナーゼ経路の構成因子であるSPS1とキナーゼドメイン間で約50%の高い相同性を示し、現在までに知られている哺乳類のMAPキナーゼ経路とは異なる新しい経路の存在が示唆された。MUK2は、低分子量G蛋白質Rac1、Cdc42の結合により活性化され、酵母の接合に関与するキナーゼSTE20の哺乳類ホモローグと考えられるPAKと同一であることが判明した。また、COS細胞に高発現させたPAK/MUK2は、MEKの活性化を通してMAPキナーゼを活性化することを示唆する結果も得た。MUK1は、STE20、PAK/MUK2ともキナーゼドメインで45%程度の相同性を示すことから、STE20、SPS1、MUK1、PAK/MUK2が新たなキナーゼファミリーを形成していることが示唆された。(MUK1とMUK2は、MAP kinase cascade Related Kinase という意味を表すMRK1とMRK2とにそれぞれ改称した。) 最近、ストレスに応答してJNK(c-Jun amino-terminal kinase)の活性化に至る経路もMAPキナーゼ経路の一つであることが明らかになったが、MUK3はこのJNKを著明に活性化することが分かった。今後MUK3の活性化機構、及び下流の因子の検索により、哺乳類の新たなストレス応答経路が解明されることが期待される。
