ヒトWilms腫瘍細胞株を用いたWilms腫瘍抑制遺伝子(WT-1)の機能解析

研究概要	(目的)本研究では、当初、1.無虹彩症患児に発生したWilms腫瘍より樹立したヌードマウス移植腫瘍株KCMC-WT-1よりin vitro培養細胞株を樹立し、性格付けを行う、2.1で樹立された培養細胞株に、Wilms腫瘍抑制遺伝子(候補)WT-1 cDNAを組み込んだ発現プラスミドを移入し、細胞の性格の変化を検討し、WT-1遺伝子の機能解析を行う、ことを目的とした。 (方法および結果)1.KCMC-WT-1の継代時毎に(現在継代21代)、培養細胞株を樹立を試みた。しかし、組織片植え込み法、酵素消化法のいずれにおいても、マウス線維芽細胞の増殖により腫瘍細胞の安定した増殖は得られなかった。現在、培養条件を検討中であるが、無血清培地、低カルシウム培地、浸透圧を変化させた培地などを用いた培養を試みている。2.KCMC-WT-1より細胞株の樹立が困難であったため、我々が樹立したラットWilms腫瘍細胞株ENU-T-1(Nagashima et al.,1989; Sumino et al.,1992, Nagashima et al.,1992)をrecipient細胞として用いることを検討した。本細胞株はノザン法にてラットWT-1遺伝子転写産物を欠いており、本実験に適している。 3.WT-1遺伝子の発現プラスミドとしてpCAGGS-neo-WT1(秋山徹博士より供与)を用いENU-T-1に、リポフェクチン法による遺伝子移入を行った。現在、得られたクローンの性格(増殖性変化、ヌードマウス移植性の変化など)を検討している。(付記)KCMC-WT-1は継代中に、組織学的に、器官様成分が失われ、腎芽細胞成分によって構成されるようになった。このことから、細胞株の樹立がより容易になると期待される。
