細胞膜結合型増殖因子変換酵素,セクレターゼの遺伝子発現クローニング

研究概要	我々が、近年同定したヘパリン結合性EGF様増殖因子(HB-EGF)は、EGFファミリーに属し、膜結合型分子(pro型)として合成され、細胞表面に発現した後、プロセッシングを受けて分泌型HB-EGFとなり、パラクリン因子として作用する。このプロセッショシングを司る酵素セレクターゼの同定を目ざして、まずその特性について検討し、以下の結果を得た。 1)HB-EGFセクレターゼの活性化は、プロテインキナーゼを介する経路で制御を受けている。 2)HB-EGFセクレターゼは、proHB-EGFの細胞内ドメインを知識し、細胞外領域の-pro^<48>-Val^<149>-部位を切断する。 3)proHB-EGFプロセッシングは、低分子量G蛋白質rhoの阻害剤であるボツリヌスC_3酵素によって阻害を受けることから、このプロセッシング機構にrhoが関与していることが示唆される。 4)HB-EGFセクレターゼは、HB-EGFをジャクスタクリン因子からパラクリン因子へと効率よく変換する。現在、HB-EGFセクレターゼの蛋白質の同定と遺伝子の単離を試みている。
