細胞増殖に関わるPKC分子種の同定とその作用機序の解析

研究概要	細胞内情報伝達系において重要なSer/Thrリン酸化酵素であるプロテインキナーゼC(PKC)は、多数の関連分子からなる遺伝子ファミリーを形成している。PKCは細胞の増殖や分化の制御に関わっていることが示唆されているが、その作用は細胞によって異なる。これは細胞毎に発現しているPKC分子種の種類が異なり分子種により生理機能が異なることに起因すると思われる。G0期のラット繊維芽細胞3Y1に増殖刺激を施すと、内在性PKC分子種のうちδとεが強く活性化されるがαの活性化は検出できないことを申請者はこれまでに報告している。そこで本年度は3Y1細胞の増殖に関わるPKC分子種を同定し、その作用機序を明らかにすることを目的とし、PKCα、δ、εの高発現株を作製し増殖刺激に対する応答を解析した。即ち、PKCδ及びεの発現ベクターとネオマイシン耐性遺伝子を3Y1に導入後ネオマイシン培地で選択し、各PKC発現株を単離した。各クローンのPKC発現量は特異抗体を用いたウェスタンブロッテイング及び細胞の[^3H]PDBu結合能の測定により求めた。次に、血清飢餓により静止期に同調したPKC高発現株をTPAや血清、EGF、LPA等で刺激し、[^3H]Thymidineの取り込みを測定することにより誘導されたDNA合成量を求めた。全てのδ高発現株は内存性のδに比べ発現量が究めて低く、顕著な効果が見られなかった。しかしベクターのみを導入したコントロール株に比べ、ε高発現株ではEGF刺激に対する応答性が若干上昇していた。そこで、より高発現の効果を検討するためにtransientな発現系でDNA合成能の解析を行う系を確立した。即ち、PKCとマーカー遺伝子(CAT)を細胞に導入しマーカーに対する抗体で細胞染色によりPKCを発現した細胞を同定し、同時に細胞にBrdUを取り込ませ抗BrdU抗体による二重染色でDNA合成の有無を判定するものである。これを用い、現在εの効果について検討中である。
