研究概要	RbはE2F、MyoD、Myogeninなどの転写因子複合体中に検出でき、CATアッセイなどにより転写活性がRbの発現により変化することから転写因子制御であると推定されている。しかし、Rbが転写因子の制御だけではなく、DNA複製に直接関与している可能性が推定される。Rbが複製に直接関与していることかどうかを検討し、癌抑制遺伝子の不活化による癌化機構の解明を行う目的で本研究を行った。これまでの研究で、細胞にc-mycプロモーター領域の複製起点を含むプラスミドをトランスフェクションした後のコピー数の変化、およびメチル化プラスミドをトランスフェクションして複製するとメチル化が解除される現象を利用して解析してきた。この結果からRbがDNA複製に関与することを強く示唆する結果が得られた。しかし、RbがDNA複製に関与する直接的な証拠とするにはまだ不十分であると評価した。そこで、複製の活性を直接検出するin vitroの系を開発して直接的な証拠を得たいと考え、Rbの免疫沈降複合体にヘリケース活性やDNAポリメラーゼ活性などがないかを検討した。ヘリケース活性、DNAポリメラーゼ活性、Rbを含む細胞抽出液からRbの免疫沈降複合体を調整した。そこにc-mycプロモーター領域を含むプラスミドを入れ、ヘリケース活性の結果生じるDNA切断点をアイソトープラベルし、Rbが存在する場合と存在しない場合、トボイソメラーゼ阻害剤存在、非存在下で比較したがDNA切断点の変化は認められなかった。また、Rb免疫沈降複合体にDNAポリメラーゼ活性も認められなかった。これまで高等動物で開発されているin vitroの完全複製系はLT複製起点を用い、LT抗原依存性の系だけであるが、LT抗原はRbの機能を抑制するために本研究においては不適当である。今後、Rbの複製への関与が直接示す目的でLT以外の複製系の開発が期待される。
