移植前ドナー抗原胸腺内投与における移植肝の生着延長効果:抗原提示細胞(樹状細胞)とTH2T細胞の役割

研究概要

1.移植肝の浸潤細胞の解析では、CD4^+T細胞には2種類のCD45RC^-CD4^+細胞とCD45RC^+CD4^+細胞が存在し、CD45RC^-CD4^+細胞にはIL-4及びIL-10のmRNAが誘導されており、これらの細胞はTh2細胞であることが判明した。
2.サイトカインの分泌パターンによりTh1とTh2細胞の区別がなされてきたが、ラットでは、leukocyte common antigenの一つであるCD45RCの発現の有無によりTh1とTh2細胞の判別が可能であることが判明した。
3.ドナー抗原投与群では、拒絶群に比較して移植肝内に多数のCD45RC^-CD4^+T細胞が認められた。しかし、脾臓やリンパ節ではこのような細胞は全く認められなかった。
4.ドナー抗原投与群では、拒絶群に比較して血中hepatocyte growth factor(HGF)濃度は有意に高かった。
5.抗HGF抗体を用いた免疫組織染色では、移植肝の門脈領域にHGF陽性細胞が認められた。また、脾臓の赤脾髄にHGF陽性細胞が認められた。
6.免疫組織多重染色法でHGF陽性細胞はHGF^+ED1^+細胞とHGF^+ED2^+細胞の2種類のマクロファージより構成されていた。
7.ドナー抗原投与群では高HGF血症を呈するが、このHGFがどのような役割を果たしているかについて検討した。migration assayではHGFがTh2細胞の遊走を増強した。
8.CD8^+T細胞のphenotypeについてFACS解析すると、CD45RC^+CD8^+T細胞とCD45RC^-CD8^+T細胞が存在することが判明した。ドナー抗原投与群では、拒絶群に比較してCD45RC^-CD8^+T細胞数が有意に増加した。CD45RC^-CD8^+T細胞はIL-4やIL-10のmRNAを発現していることが報告されており、ドナー抗原投与群ではTh2タイプのCD8^+T細胞が多数浸潤していることが判明した。
以上の結果より、ドナー抗原投与群ではCD45RC^-CD4^+T細胞やCD45RC^-CD8^+T細胞のphenotypeが有意に増加しており、免疫学的寛容状態の誘導の機序のひとつであると考えられた。
