研究概要

7年度は、1)ADF結合蛋白質の単離、およびその機能解析、2)ADF遺伝子発現調節部位結合因子の同定、単離、3)ADF欠損マウスを作製し、これを用いて、個体レベル、もしくは種々の組織から細胞単離し、正常及びADF-transgenic mouceとの比較において、ADFの抗酸化機能について解析を行うこと、4)核内において、及び、NF-kB,thioredoxin/ADFなどの蛋白性因子の還元状態保持に寄与する物質の単離、同定する事などを目指し、その為に必要な研究交流・情報交換を行うことが目的であった。
7年度においては、各プロジェクトは総体的に、順調に進んだ。具体的には、1)ADF遺伝子プロモーター部位のなかで、酸化ストレスに応答する領域の特定を行った、2)ADF欠損マウスの作成に成功した、3)ADF結合性蛋白質のcDNA単離が既に進行し、幾つかの候補cDNAを得た、等が挙げられる。
特に、同定したADF遺伝子プロモーターの酸化ストレス応答領域の核酸配列は、他の既知の制御領域配列とは異なっており、新しい制御因子の関与が示唆され、ADF結合性蛋白質のcDNA単離の結果、従来ある程度予想はしていたが、ADFがマクロファージなどのoxygen burst過程に関与している可能性が示唆された。
