南米先住民族の人類遺伝学的研究

研究概要

アンデス先住民族(アタカマ族)には南西日本に多いヒト白血球ウイルス(HTLV-I)感染者が高頻度(10%)に出現し、しかも彼らは南西日本人と類似したHLA遺伝子を有しており、両者の遺伝学的背景やがん罹患分布には類似性が見られ、民族疫学的にも興味が持たれている。また、チリ北部の砂漠地帯には現在でも数千検体のミイラが比較的好条件で残存している。そこで、今年度はこれら先住民族のミイラを保存しているサンミグエル博物館(アリカ市)やサンペドロ博物館、およびミイラが未だに埋葬されている同地域の古代墓地を訪問し、同博物館の考古学者らの協力を得てミイラ138体(アリカ市96体、サンペドロ市42体)から組織(骨、および骨髄)を採取することがきでた。それらの組織はDNA(核、ミトコンドリア)を検索するために日本まで持ち帰った。まず、サンペドロ博物館の約千年前の標本14検体につき、乾燥量100-500mgを用い、PCR法にてヒト白血球抗原と血色素のDNA断片の抽出を試みた。その結果、4検体からHLADQB1(400bp)とβクロビンDNA(800bp)が得られた。これらの結果を踏まえて、さらに他の検体からのDNA断片の抽出作業を続け、それら抽出したDNA断片の配列を明らかにしていきたい。また、チリ先住民族や日本人に多く検出されているHTLV-IのプロウイルスDNAの抽出も試みたが、現時点ではその抽出には成功してない。ミトコンドリアDNAの抽出にも成功し、その配列について検討中である。次年度も今年度に引き続き、ペル-南部やチリ、ボリビア北部のミイラから検体採取を続け、それらの核DNAやミトコンドリアDNAの抽出を続けていく。それらのDNA情報と現存しているチリ先住民族や日本人のDNAとの比較研究により、民族疫学的、免疫遺伝学的、人類学的、考古学的に有用となるDNA情報として構築していきたい。
