研究概要

本研究の主たる目的は、イスラエル側のマクロファージの分子・細胞生物学的研究に関する知識技術やトランスジェニック作出に関する知識・技術の提供を受けると同時に、我が国側で、東大牧場でクローズドコロニーとして維持されているシバヤギを材料として、子宮上皮細胞、トロフォブラスト細胞、ならびに、マクロファージの培養系を確率し、胚移植後の着床効率を高めるための基礎知識を得ることにある。Ashuva Knyszynski博士の来日を仰ぎ、シバヤギの培養細胞系確率を試みた。子宮上皮細胞系については、培養細胞下部を確立することができた。現在、クローン化の実験を進めている。一方、トロフォブラスト細胞については、予備的にトロフォブラスト細胞と判定された細胞について、培養系の確率を図り、明らかに繊維芽細胞とは挙動の異なる培養細胞系を得た。なお、イスラエル側に送って実験する計画であったが、得られたマクロファージの適当な採取・培養法の確率ができぬ段階で、研究が終了することとなった。なおイスラエル側では、マクロファージの活性化におけるシグナルトランスダクションについて研究を進め、主として、ラットおよびマウスのマクロファージを用いて実験を行ない、MAPキナーゼの関与について興味深い知見が得られつつある。一方、われわれの研究室では、マウスならびにシバヤギを用いて、胎盤に置けるホメオボックス遺伝子群の発現動態の解析、マトリックスメタロプロテアーゼcDNAに関する研究を行ない、興味深い知見を得ることができた。さらに、マウスを用いた卵成熟の研究で著名な、Nava Dekel博士に来日を依頼し、家畜における胚操作の一環として重要な、invitro卵成熟におけるシグナルトランスダクション系の役割に関する最新の研究成果や技術の情報を得ると共に、Weizmann研究所で所有するMRI装置によるラット着床部位や時期の非破壊的検出と、その着床・胎盤形成研究における応用について、今後の共同研究の可能性を論議した。また、イスラエルでトランスジェニックヤギ作出に成功しているMoshe Shani博士には、実際のトランスジェニックヤギ作出の作業を見学させてもらうことができた。また、博士に来日を依頼して、最新の研究情報を取得するとともに、今後の相互の研究交流の端緒を開くことができた。今回の共同研究は、東大牧場のシバヤギの繁殖率の低下、Weizmann研究所における研究体制の大幅な改組など、不足の事態が生じ、当初予定sた研究計画を消化することが困難であったが、今回の研究を端緒としても、現在も、両サイドで研究を研究を継続しており、今後、さらに、重要な成果が、日本、イスラエルの双方で挙げられることが期待される。
