研究概要

タンパク質の熱物性と構造・機能の関係を明らかにする目的で、イソプロピルリンゴ酸デヒドロゲナーゼをモデル酵素に選び、物理科学的な解析を国際協力して行った。日本側はキメラ酵素を中心に安定化変異体と機能改変酵素の作成、それらの結晶構造解析を分担した。また、国外共同研究者に酵素の精製品を提供した。アメリカは大腸菌、サルモネラ菌の酵素の三次元構造解析、機能改変酵素の作成、及び低温の結晶構造の揺らぎの解析を分担し、ハンガリーは大腸菌の酵素の構造を予測し、安定化設計を行った。大島と田中はアメリカ側の研究室を訪問し、変異体の調整と共同論文の作成を行い(大島)、また、低温下の結晶構造解析を実験した(田中)。アメリカ側研究分担者は大学院生(Gerlind Wallon)を日本に派遣し、大腸菌変異体(アメリカにおいて調整)の熱物性の測定を日米共同で行った。ハンガリーの研究分担者は、本研究費補助金とは別の独自の経費を用いてアメリカ側研究分担者を訪問し、共同実験を行ったが、日本ーハンガリー間はデータの交換による共同研究に留まった。
国際共同研究の結果、大腸菌のイソプロピルリンゴ酸デヒドロゲナーゼの立体構造解析を完成した。また、極低温下結晶構造解析の測定法を開発し、予備的な測定に成功した。高度好熱菌の生産する耐熱性イソプロピルリンゴ酸デヒドロゲナーゼではループ部分が強固となっていることが明らかとなった。これに基づいて、大腸菌の相当する酵素の耐熱下が設計され、変異体酵素が作成された。しかし、予測に反して安定化されたいなかった。一方、進化分子工学手法を用いて、常温適応した耐熱性酵素をランダム変異体から選択する系は日米が各独自に開発を終わり、日本側はすでに改変酵素の取得に成功した。構造解析は次年度の課題である。アメリカ側はさらに系の改良が必要である。
