研究概要

2年目の本年度には昨年度の相互訪問の成果が現れ、植物の個体生長と細胞機能の制御の分子生物学的解析に取り組むNUとUCBの研究者の間で、それまで交流のなかった者の間でも電子メール番号の交換などによって、頻繁に情報交換、研究試料やプロトコルの交換が行われた。NUの中村、杉山、前島はUCBのJonesとCa^<2+>-情報伝達系や液胞機能を中心に活発な情報交換を行い、中村はタバコから糖誘導性の特異な細胞膜結合型カルシウム依存性プロテインキナーゼを約1,000倍に部分精製し、杉山は窒素同化系遺伝子の発現制御におけるCa^<2+>-情報伝達系の関与を明らかにした。前島とJonesはCa^<2+>-トランスポーターやCa^<2+>チャンネルに関する研究を進め、前島は後者のcDNAを同定し、Jonesは急速冷凍法による顕微鏡観察によりオオムギ種子のアリューロン層の細胞内に多様な液胞や小胞様のユニークなオルガネラを見出し、前島と共同で液胞膜特異タンパク質に対する抗体を使った研究を進めた。NUの杉浦とUCBのGruissemは葉緑体遺伝子のin vitro発現系に関して活発な情報交換を行い、杉浦らは葉緑体抽出液によるin vitro翻訳系を、またGruissemらはpre-mRNAプロセシング系を開発した。昨年に引き続き、シロイヌナズナを使った分子遺伝学的解析の情報交換と研究材料の交換が行われ、UCBのFischerはT-DNAタッギングラインからの胚発生に関する突然変異の解析を進め、NUの町田は生長速度が著しく遅くなった変異株を単離して原因遺伝子を同定した。相互派遣計画は多少の変更を余儀なくされたが、頻繁な情報交換によって研究は順調に進行した。年度末のために本助成枠外になったが、UCBのBuchanan,Gruissem,Fischer,Jones,Lemauxが日本植物生理学会大会シンポジウムの演者として来日する機会に、NU側研究者と本研究の総括と、共同研究を一層発展させるための今後の方策について討議する予定にしている。
