研究概要

AIDSは、通常のウイルス感染症とは異なり、ヒト免疫不全ウイルス(HIV)の感染後、平均10年近い無症候性キャリア(AC)期を経た後に初めて発症する。従って、従来のHIV研究の中心課題であったHIVの複製機構を基盤として、その潜在機序、さらに潜在ウイルスの活性化機序を明らかにすることが発症病態を理解する上で極めて重要である。私達は今までに、1)HIVの持続感染成立には、nef遺伝子が正常でvif、vpr、またはvpu遺伝子の変異が必須であること、2)nef遺伝子産物が、免疫担当細胞への増悪因子としても働き得ること、3)感染初期において、nef遺伝子産物に対する細胞性免疫の方が、env遺伝子産物に対するよりも、ウイルス負荷の制御に効果的に機能していること、4)不完全粒子によるアポトーシス誘導が起こることを突き止めた。そこで、本研究では、AC期のHIV潜在機序、さらにその潜在状態からAIDS発症への移行過程に認められるウイルス活性化機序に関して、次の研究を行った。
1)アクセサリー遺伝子の内、vifまたvprの変異、またはvifからvprにまたがる欠失(ミスアライメント機序)が、持続感染成立に重要であることを見い出した。
2)nef遺伝子の産物(可溶性Nef)の潜伏HIVの活性化の機序として、MOLT-4細胞表面に35kDaのNefレセプターと考えられる分子を同定した。また、このNef結合に伴ない、Ash/Grb2により30kDa分子がリン酸化され、更にNF-kBが活性化されることを明らかにした。
3)有意にNef蛋白合成を抑制し得るnefアンチセンスDNAを開発した。
4)不完全(pol proteaseとnefにnonsense変異、env gp41にアミノ酸置換がみられる非感染性)HIV-1粒子が、HIV-1感染生体内でみられるようなバイスタンダー細胞のアポトーシス誘導をおこすことを見い出した。同様なプロテアーゼ変異を、米国のHIV-1感染者の末梢血中のHIV-1の中に見い出した。また、タイのHIV-1感染者由来臨床分離株の中に、同様のアポトーシス誘導能のあることを見い出した。
5)日和見感染症としてのボルナ病ウイルスの活性化を、タイHIV-1感染者を対象として行い、HIV-1 E型感染STD患者に有意に上昇していることを見い出した。
