研究概要	複製・転写調節因子MSSP‐1およびMSSP‐2について、細胞内でC‐MYC、cdcキナーゼを含む複数のタンパク質と複合体を形成すること、さらにin vitroでDNA複製の主体酵素であるDNAポリメラーゼαと直接結合し、同酵素活性を促進することが昨年度に判明し、MSSPとの複合体形成において、C‐MYCはcdcキナーゼやDNAポリメラーゼと競合し、細胞周期のG1期からS期への進行を制御・確認する、いわゆるライセンス化を行っていることが強く示唆された。MSSP自身の転写も、MSSPゲノム遺伝子の解析から、転写プロモーター上流配列により細胞周期依存的に調節されており、C‐MYCと同様にG1初期に発現が誘導されて中期からS初期にかけてピークに達することが明らかになった。MSSPを細胞に導入・強制発現させると、複製だけでなく種々のプロモーターからの転写が活性化される。そのため、MSSP.は複製・転写の両方の調節に関わる囚子であると考えてきたが、主として複製開始の調節に直接関わり、C-MYCと共に細胞をS期ヘとライセンス化し、その結果として関連遺伝群の転写が促進あるいは抑制されることが示唆された。また、酵母系を応用したスクリー二ングによりC-MYC結合タンパク質として新たにクローニングされたAMY‐1(associated with Myc)はMSSPと同じくC‐MYCのN末端付近のmycbox配列を含む領域を認識・結合した。AMY‐1は転写促進活性を有し、通常は細胞膜に沿って細胞質側に局在しているが、C‐MYCの発現が高まるG1後期からS期にかけて核内へ移行した。複製囚子MSSPと転写因子AMY‐1のC‐MYCへの競合的結合はC‐MYCによるS期ライセンス化の制御スイッチの役割を果たしていることが強く示唆された。
