研究概要

「エキノコックス症、襄虫症の血清学的鑑別診断法に関する研究」が平成6年度単年度の国際学術研究として採択されたのに引き続き、平成7年、8年度の国際学術研究として「難治性寄生虫病(エキノコックス症、襄虫症)の分子・免疫生物学的研究」が2年継続研究として採択された。平成7年度の国際学術研究では研究代表者、伊藤亮が最近報告した(a)Em18抗原の種特異性、診断用抗原としての有用性についての評価、(b)Em18抗原を遺伝子工学によって作製する試み、(c)襄虫症診断の現状、(d)伊藤が最近発見している低分子抗原の有用性の評価を中心とする共同研究を主な目的とした。以下にその実績概要を報告する。
(1)多包虫症患者における病態と抗体応答との相関についての解析:中国、北西部における単包虫症、多包虫症両症が流行している地域住民の抗体応答を解析した結果、Em18に対する抗体応答は多包虫症患者に特有であること、特徴的なIgGサブクラスはIgG4であることが判明した(Wen et al.1995,Ito et al.1995、その他)。(2)Em18ウェスタンプロット法による多包虫症と襄虫症者との鑑別診断の可能性についての解析:等電点電気泳動法によって分画した多包虫、襄虫抗原を用いてEm18の種特異性を確認し、同時に幾つかの襄虫症特異抗原を発見した(論文作成中)。(3)Em18-ELISA法の確立:等電点電気泳動法によって分画した部分精製抗原を用いたPP-Em18法とEm2-ELISA法とを比較解析しPP-Em18-ELISA法の信頼性が現在WHOが推奨しているEm2-ELISA法よりもその特異性において優っていることが判明した(Ito et al.Lancet投稿中、その他)。(4)Em16遺伝子組み換えEm16のクローニング:クローンを無くして困難をきたしていたEm16に対するモノクロナル抗体を作製し、原頭節から作製した多包虫cDNAライブラリーを用い、これからEm6の遺伝子クローニングを始めるところである。現在、Em18に対するモノクロナル抗体作製を試みている。
