研究概要	難治性寄生虫病(エキノコックス症、曩虫症)は、近年世界各地で流行が拡大しており、WHOの新興感染症についての最新の報告書Emerging and other communicable diseases:strategic plan 1996-2000(WHO/EMC/96.1)においてもHIV/AIDS,Ebola,dengue,drug resistant bacterial infectionsその他と一緒にechinococcosis,zoonosesとして取り上げられている。本年度の研究目的は昨年度に引き続き、これらの寄生虫病(多包虫症、単包虫症、嚢虫症)についての血清診断法の確立である。多包虫症についての血清学的鑑別診断法の開発として、等電点電気泳動法により多包虫症に特異的なEm18を主成分とする分画の作製を試みた。これまでのところEm18とEm16とが混在した分画が得られているだけであるが、この部分精製抗原を用いたイムノブロット法、ELISA法を開発し、WHOによって推奨されているEm2plus-ELISA法との間での信頼限界についての相対評価を試みた結果、我々の方法がより高い信頼性を有していることが判明している(Ito et al.1997,Clin.Diag.Lab.Immunol.4,57-59,Ma et al.1997,Trans.Roy.Soc.Trop.Med.Hyg.in press,Ito et al.1997,Jpn.J.Med.Biol.Sci.inpress)。単包虫症については多包虫抗原を用いたイムノブロット法によりEm18に対する抗体応答の欠損に基づいて判定する方法が最も信頼性が高いこと、外科的に摘出された病巣を検査材料として多包虫、単包虫の遺伝子鑑別診断が可能なことが判明している(Ito et al.in prep)。嚢虫症についても、これまでの方法とはまったく別の方法によって非常に特異性が高い抗原成分をイムノブロット法によって検出する方法を開発している(Ito et al.in prep)。以上の成績から世界規模で流行が年々深刻化してきている新興感染症、エキノコックス症、嚢虫症についての血清診断法が基本的に確立できたと結論する。今後の研究課題は、特異抗原を遺伝子組換え抗原として作製すること、遺伝子診断法の簡略化を試みること、流行地域での疫学研究に応用することである。
