研究概要

ヒトのヌクレオチド除去修復機構(NER)の初期過程に必須の色素性乾皮症A、C、G群、コケイン症候群B群の原因遺伝子(それぞれXPA、XPC、XPG、CSB遺伝子)、およびERCC1遺伝子等の機能の解析を行った。田中とWoodは、XPA蛋白質が損傷DNAに選択的に結合する損傷認識蛋白質である事を明かにし、そのDNA結合ドメインが、Znフィンガーモチーフを含む122個のアミノ酸領域にある事を見いだした。さらに、田中は、XPA蛋白質がERCC1蛋白質と結合し、その事でXPA蛋白質の損傷DNAに対する結合能が上昇する事を、Hoeijmakers、安井やWoodとの共同研究で明かにした。この相互作用が損傷DNA認識過程に関与する事を示唆する。WoodとHoeijmakersは、ERCC1蛋白質がXPF (ERCC4)と複合体を形成し、損傷部位の5′側でDNA1本鎖切断を行うエンドヌクレアーゼ活性を担う事を示し、XPF遺伝子のク-ニングにも成功した。Woodは、replication protein A (RPA)がNERの初期過程に必須である事を示していたが、田中はXPA蛋白質がRPAと結合し、損傷DNAに対する結合能が相乗的に上昇する事を明かにした。Woodは、XPG蛋白質が損傷部位の3.′側でDNA1本鎖切断を行うエンドヌクレアーゼである事を見つけたが、XPG蛋白質がRPAと結合する事も解かった。これらの結果はXPA/ERCC1/XPF、XPA/RPA/XPGの相互作用により、各エンドヌクレアーゼが損傷部位に誘導される機構が示唆された。花岡はHoeijmakersと共同し、XPC蛋白質がRAD23ホモログと結合し、それがNERに必須である事を示した。Hoeijmakersと田中は共同し、XPA/CSBダブルノックアウトマウスを樹立し、その神経症状の有無について解析している。
