超活性海洋天然物の精密構造解析

研究概要

シテガラ関連毒として発見され、最も複雑、かつ猛毒とされるマイトトキシン(C_<164>H_<256>O_<68>S_2N_2)の推定構造をさらに確実にするために、三次元NMRを測定し、^<13>Cおよび^1Hの全シグナルの同定と環状エーテル骨格部の相対立体配置を決定した。また、この巨大な分子をフラグメントに分割することなく、MS/MSスペクトル測定に供し、構造確定に成功した。環状構造間に存在する鎖状部は合成品とのスペクトルを比較することで決定した。関連して発見された渦鞭毛藻Ostreopsis siamensisの生産する毒を、パリトキシンの新規類縁体(bisdemethyldeoxypalytoxin)と同定し、パリトキシンの起源を解明した。
世界の各地で大きな被害をもたらす赤潮生物プリムネシウム・パルバムの毒の主成分であるプリムネシン-2の単離に初めて成功し、その平面構造を決定した。本化合物は分子量が1968で、分子式C_<96>H_<136>Cl_3NO_<35>を有する巨大分子である。分子内に14のエーテル環と16の水酸基、アセチレン基、共約二重結合.塩素原子、アミノ基などの多彩な官能基を有し、炭素原子90で構成される長大な炭素鎖を骨格とする。天然有機化合物としては前例のない、特異な構造である。魚毒性や溶血性も強力であり、特に溶血性は植物サポニンの7千倍にも達する。
ニュージランドで発生した神経性貝毒の原因毒として、ブレベトキシンの新規誘導体群の構造を決定した。エーテル環の解裂や極性基やアシル基の付加を伴うこれらの誘導体の構造は、従来提案されていたブレベトキアインの作用モデルを破棄し、あらたなモデルの構築を迫るものである。関連して、新規の大炭素環イミンであるギムノジミン(C_<32>H_<45>NO_4)の構造を決定し、この毒の起源が渦鞭毛藻のGymnodinium sp.であることを確定した。
その他、テトロドトキシンの新規同族体、オゴノリ毒ポリカバノシドの新規同族体群の構造も決定した。また、オカダ酸の放射性標識体を調製し、タンパク質脱燐酸酵素2A型酵素との相互作用を解析した。
