大脳認知記憶の神経機構:機能的磁気共鳴画像法と細胞レベルアプローチの統合

研究概要

1.機能的磁気共鳴画像法(fMRI)を用いて高次機能の大脳内局在を解析する研究が進展した。fMRIの利点である時間分解能を利用して"trial-base"の解析を行う方法を開発し、GO/NOGO課題のNOGO試行に特異的な活動部位を前頭葉BA44/45野に同定した。この部位は使用する手によらず右半球優位をいう特性をもっていた。さらに、ヒト前頭葉症状の診断に最も頻繁に使用される課題である"カードソーティング課題"をコンピュータ化し、fMRIの"trial-base"解析を適用する方法を開発した。
2.エコープレーナー(EPI)磁気共鳴画像技術実用化の目標はほぼ達成された。MRI制御システムを従来の専用ハードウェア型から汎用ワークステーション型へ改造工事はやや遅れ、本年8月になる見込み。これにより、さらにマルチタスク制御性能の向上、メモリ・ディスク管理機能の強化等が達成される。
3.サル部分分離脳標本においてサッカード対連合課題データを収集し、半球間転移をおこさない情報とそうでない情報を特徴ずけた。完全分離脳標本などのコントロール実験や蛍光トレーサーによる分離の確認実験などを追加し、前頭葉における視覚信号の特性を明らかにした。
4.RT-PCR法、RNAase protection法 並びに northern blotting法をサル大脳 mRNA量の定量に適用するプロトコルを開発し、サル大脳(ことに視覚連合野・辺縁系)各部のNeurotrophinファミリー、trkファミリーの定量を行った。RT-PCR法の内部標準の取り方を工夫してnorthern blottingの結果と高い相関を得ることができ、northern blottingの適用できない微小領域(例えばBA36野)の遺伝子発現計測に有用であることを立証した。霊長類腹側視覚経路に沿って、とくにBDNFとTrkBが高次情報処理と相関するグラディエントをもつ事を示した。
