大脳認知記憶の神経機構:機能的磁気共鳴画像法と細胞レベルアプローチの統合

研究概要

1.エコープレーナー(EPI)磁気共鳴画像技術実用化の目標はほぼ達成された。このシステムを用いてEPI磁気共鳴画像法の利点を生かした事象関連型解析法(event-related fMRI)を開発し、ウィスコンシンカード分類課題において活動する脳部位を同定した。この課題における注意のシフトを実現する脳活動は、以前に報告したGO/NOGO課題のNOGO試行に特異的な脳活動部位と極めて近い部位であることが判明したので、実際に部位的重複があるかどうか検討した。その結果、二つの課題に対する脳活動は、ピークとなるピクセルが完全に一致し統計的に有意な重複が存在した(Brain 122,981-991,1999)。さらに、注意のシフトを実現する脳活動は作業記憶課題においても重要な役割を果たす事が予想されるので、作業記憶課題との関連を検討したところ、二つの課題における活性部位に有意な重複が存在することが明らかになった(Cerebral Cortex 9,745-743,1999)。
2.前頭葉から側頭葉へのトップダウン信号の存在を証明し、その性質を解析するのに成功した(Nature 401, 699-703,1999)。サル部分分離脳標本を用いた電気生理学的解析によって、視覚対連合記憶課題において、前頭葉から側頭葉への逆向性信号が存在し、課題遂行に重要な役割を担っている事を発見した。トップダウン信号の潜時はボトムアップ信号の潜時より約100ミリ秒長く、前頭葉における情報処理に要する時間に対応すると考えられた。また、トップダウン信号は刺激の物理的特性よりむしろその意味的性質をコードしており、遅延期においては想起すべき対象に関連した信号を運んでいることも明らかになった。さらに、このトップダウン信号が皮質下の経路ではなく、皮質間連絡繊維により直接運ばれることも証明した。
