新規不斉連続反応の開拓と生理活性天然物の効率合成への利用

研究概要	不斉分子内二重ミカエル反応及び不斉分子内ミカエル・アルドール反応の確立を目的に不斉環境下での反応を検討した。先ずキラル補助剤を反応系に組込むことにより、ほぼ100%deのもとで不斉分子内二重ミカエル反応を達成し得た。なお本化合物より制ガン性アルカロイドの光学活性チロホリンの全合成を完成した。本研究によりチロホリンの絶対配置を決定し得た。しかしながらキラル塩基存在下での反応では、最高約50%eeを示す連続反応成績体を得るにとどまっている。次いで分子内二重ミカエル反応をβ位にフェニルスルフェニル基をもつエノン系に適用したタキサン骨格の合成を試みたが、目的物を得ることはできなかった。また桜井・細見反応を分子内反応として活用しタキサン骨格形成に付したが、新しい型の分子内ドミノ型反応が進行し高歪系多環性化合物が生成した。一方スルホンを活性化基とする分子内アルドール反応を試みた場合、タキサン骨格のBC環部に相当するビシクロ[6.2.0]ドデカンをほぼ定量的に合成することに成功した。本知見は制ガン剤タキソ-ル合成の有力なルートを提供している。
