種々のサイトカインのレクチン様活性とその生理的意義に関する研究

研究概要	種々のサイトカインのレクチン様活性は、生体内におけるサイトカインと標的細胞の間の認識機構およびそれぞれのサイトカインのクリアランス機構を考える上で重要である。我々はTNF-α、リンホトキシン(LT)、IL-1βが類似したレクチン様活性を持っており、これら三者がGPIアンカーのグリカン部分を認識していることを示唆するデータを得ていた。本年度は[^<35>S]メチオニンでラベルしたrecombinant IL-1βを調製し、詳細な糖結合特異性を調べた。 1.[^<35>S]メチオニンラベルしたIL-1βの調整法ヒトIL-1βのcDNAをpET3プラスミドのT7RNAポリメラーゼプロモーター下流に導入したものをテンプレートにしてTNT Coupled Reticulocyte Lysate Systemsの系に従い、[^<35>S]メチオニン存在下においてin vitroの転写翻訳を行わせた。4μgのDNAプラスミドテンプレートから500fmoleの[^<35>S]IL-1βが得られた。 2.固定化糖蛋白質への[^<35>S]IL-1βの結合プレート及びカラムに固定化した各種糖蛋白質のうち、C-末端にGPIアンカーのグリカン部を有するヒト胎盤アルカリホスファターゼ、ウロモジュリン、トリパノゾームVSG、癌胎児性抗原が[^<35>S]IL-1βと結合した。これらの結合はGPIアンカーの部分構造を担うマンノース-6ーリン酸やAc・NH・CH_2・CH_2・O-HPO_3→6Man-propylやAc・NH・CH_2・CH_2・O-HPO_3→6Manα1→2Man-propylで阻害されることから、IL-1βと糖蛋白質の結合はGPIアンカー部糖鎖を介していることが示唆された。 3.極性化したMDCK細胞への[^<35>S]IL-1βの結合生体の各種臓器の上皮細胞は極性化しており、頂端部側にGPIアンカーした糖蛋白質が配列していることが知られている。MDCK細胞(イヌの遠位尿細管部の上皮細胞)をフィルター上で極性化して培養し、頂端部側と基底部への[^<35>S]IL-1β結合活性を測定した。その結果、頂端部側のみIL-1βが結合する事から、IL-1βはGPIアンカーのグリカン部を介して細胞に結合することが判明し、その生理的意義を解析中である。
