研究概要

40〜35億年前の太古の地球で、無酸素下に誕生した2タイプの緑色と紅色光合成細菌の融合で初めて1つの細胞内に2タイプ(系I-系II)の反応中心をもつシアノバクテリアが生まれた。我々はこれらの反応中心の電子移動機構の解明、比較、さらに始源的な反応中心の可能性の探査をおこなった。反応中心複合体では蛋白質は反応分子の配置・配向を固定するだけでなく、エネルギーレベルや反応経路を制御することを明らかにした。本研究では、植物光化学系I反応中心のphylloquinone(ビタミンK1)を別の化合物では置換し、キノン反応の△G依存性、電子移動における蛋白質の役割、反応最適化機構を検討した。この結果蛋白質の変異が反応最適化をもたらしたことがあきらかとなった。絶対嫌気性の緑色細菌についても研究をすすめ、この細胞の反応中心はユニークな植物と細菌型の2種のクロロフィルをもち、チトクロムも独特な膜結合性のものであることを示した。光化学系2の酸素発生型反応中心でもキノンの入れ替えを行い、その特徴をあきらかにした。またこれまでに知られていなかったZn結合型クロロフィルをもつ細菌を発見し、その色素系電子伝達系の特製の解明を通じて、光合成生物の進化と古地球環境の相互作用がこれまでとはまったく異なる視点から考察できることをしめした。
