研究概要	本研究プロジェクトは、自己認識T細胞クローンの選択的分化機構を分子レベルで解明することを目的としている。昨年までの研究では、この解析に有用な系としてTCRジェニックマウス(TCR-Tg)を作成し、T細胞レパートリー形成のための正・負選択には2段階あることを示した。本年度はTCR-Tgを用いて、positive selection (P.selection)の基本的機構であるTCRを介したシグナルとその発現調節に関して以下の事柄を明らかにした。 (1)P.selectionを受けたりDP未熟胸腺細胞はTCR発現を維持するが、刺激を受けないDP未熟胸腺細胞ではTCR発現は低下していく。DPステージに於けるTCRの発現低下現象は、CD4とMHCの相互作用によるととの報告があるが、今回の研究ではTCR発現低下はTCR-MHC相互作用により阻止されることを初めて証明した。さらに、CD45欠損マウスとTCR-TgのFlを作製して、DP細胞におけるTCR発現の抑制はCD4-lckシグナルにより誘導され、一方、TCRシグナルがその伝達系に加わると、その抑制シグナルが阻止されることを示した。(2)DPステージでのTCR-MHC相互作用では、両分子間のavidityが弱いのにもかかわらず、MHC提示の抗原認識をする末梢T細胞と同じく、ZAP-70、CD3-γ、PLC-γといった基質のチロシンリン酸化の誘導がin vivoでおこることを明確にした。また、P.selectionに於けるZAP-70リン酸化の必要性は、MHC classll拘束性TCRをもつDP細胞では少ないことが示唆された。
