研究概要

昨年度までにsmall Maf蛋白がCNC familyのbZip蛋白とヘテロ2量体を形成して血球細胞特異的な転写因子NF-E2として機能することなどが明らかになっていた。このNF-E2やv-Mafの転写活性化能を解析する過程で様々な細胞にMafあるいはNF-E2ヘテロ2量体とDNA認識特異性で類似する内在性の極めて強い転写促進活性の存在が観察された。この転写活性化因子は特にNF-E2と類似した認識配列特異性を有するが、gel shift法による解析ではNF-E2やMafのホモ2量体などよりかなり大きな移動度を示した。またMaf family蛋白のdominant negative mutantを細胞に発現する実験などからも、この内在性の強い転写促進活性の本体はおそらく未知のbZip蛋白のヘテロ2量体であり、しかもそのcomponentのいずれかはMaf family蛋白と2量体を形成しうる可能性が強く示唆された。そこでこの因子のcloningを試みている。現在までにWest-western法で未知の遺伝子がいくつか得られ、Two-hybrid法によるcloneも解析中である。既に明らかになっているFos, Junなどとのヘテロ2量体形成に加え、さらに多くのbZip蛋白との2量体形成を明らかにし、その多様な2量体のDNA認識特異性、発現などを明らかにすることによって、Mafファミリー蛋白の正常機能、過剰発現による細胞がん化の機構なども明らかにしていくことが出来るものと期待される。
