マウスSEP1ホモログの初期発生、配偶子形成、免疫グロブリン再構成における役割

研究概要	出芽酵母のSEP1遺伝子産物(Sep1p)は、DNAの組換え、RNA代謝、微小管形成、胞子形成、テロメア形成、細胞の老化など、細胞内の様々な機能に関与していることが知られている。本研究では、これらの細胞レベルで明らかにされている機能が、哺乳動物の個体レベルでどのように反映されるのかを調べることを目標とした。そのためにマウスのSEP1関連遺伝子のcDNAの単離を行い、そのcDNAの構造と機能を解析した。マウス精母細胞cDNAライブラリーより得られたDhm2は、出芽酵母の減数分裂や老化に重要なSEP1の相同遺伝子である。Dhm2は1687アミノ酸からなる蛋白質をコードする。本研究により、Dhm2は単に構造的のみならず、機能的にも出芽酵母のSEP1と相同性を有していること、Dhm2の発現は精巣特異的に行われていること、が明らかになった。さらに、Dhm2ホモログのDhm1が生育に必須であることがわかったことから、Dhm1の変異マウスの作成は困難となった。しかし、酵母dhp^+遺伝子の研究によってその可能性が開けてきた。酵母dhp1-1ts変異は、野生株Dhp1pのC末端領域を欠失させることによって得られたものである。従って、マウスDhm1についても、Dhm1pのC末端を欠失させることによって変異遺伝子を得、変異マウスを得るという戦略を立てることが可能になった。この研究によって、Dhm1及びDhm2のノックアウトマウスの作製を通じて、Dhm1及びDhm2の発生段階における要求性や、個体の老化、配偶子形成における役割を明らかにする基礎が築かれた。
