植物種子のジベレリン誘導性システインプロティナーゼの解析

研究概要	2種のトウモロコシ種子CPであるCCP1およびCCP2を対象にして解析を行った。しかし通常のトウモロコシの種子には内生GAがもともと高濃度で含まれているため、GA矮性トウモロコシ(d5)を用いた。まず、ノーザン解析を行ってCCP1mRNAおよびCCP2mRNAのd5種子中での発現を見ると、両者とも、GA3処理した発芽種子においてのみ発現が増加した。この場合、GA阻害財BX-112存在下では両mRNAの発現は有意に抑制された。したがって、CCP1もCCP2もオリザインα、β、γおよびアリューレインと同様にGA誘導性酵素であると結論した。しかしcDNAの解析から推定されるアミノ酸配列を比較すると、CCP1とCCP2の間の相同性は低く、42%であった。また、時期特異的発現パターンをノーザン解析で調べたところ、CCP1mRNAは登熟期にすでに存在するのに対し、CCP2mRNAは完熟種子にさえ存在せず、発芽過程ではじめて発現することが判明し、両酵素の植物生理学的役割は明らかに異なっていることが示唆された。また、アミノ酸配列を見ると、CCP1は乾燥ストレスにより発現が誘導されるシロイヌナズナ葉茎CPおよびエンドウ葉茎CPとそれぞれ79%および72%という高い相同性を示す。しかしCCP1は種子を乾燥させても発現誘導を受けなかった。このことから、CPの乾燥誘導とタンパク質構造上の類似性との間に一義的な因果関係は見いだせなかった。一方、CCP2はオリザインγおよびアリューレインとそれぞれ87%および89%の高い相同性を有した。
