研究概要

TGF-βスーパーファミリーの因子であるアクチビン、BMPは二種類のセリン-スレオニンキナーゼ型レセプター(I型とII型)によってシグナルを伝達する。本研究ではTGF-βスーパーファミリーの形態形成における役割をレセプターのレベルで明らかにするために研究を行なった。
1.アクチビン、BMPのレセプターの組織での発現
我々がクローニングしたアクチビン、BMPのI型レセプターのさまざまな組織での発現を調べた。成人ヒトではBMPR-IAは骨格筋や心筋をはじめさまざまな組織に広く見られるのに対しBMPR-IBは脳にのみ発現が見られた。胎生マウスでは肝臓を除くほとんどすべての臓器に発現が見られたが、BMPR-IBの発現パターンはBMPR-IAより限局して見られた。ラットの大腿骨を実験的に骨折させると、BMPレセプターの発現が骨折の修復に伴う骨形成のさいに誘導され、骨形成に重要な役割を果たしていることを示唆する所見を得た。
2.新しいセリン-スレオニンキナーゼ型レセプターのリガンドの同定と、新しいBMP様因子GDF-5のレセプターの研究
リガンドが同定されていなっかたセリン-スレオニンキナーゼ型レセプター(U2)がBMPのII型レセプター(BMPR-II)であることを明らかにした。またOP-1(BMP-7)はBMPR-IIだけでなくアクチビンのII型レセプターに結合し、アクチビン様の作用を持つことを示した。
GDF-5は新しいタイプのBMPで、本研究によりBMPR-IBとBMPR-IIに結合することが確認された。BMPR-IAに対する結合は顕著ではなかった。BMPR-IBの発現はBMPR-IAに比べて限局していることからGDF-5と他のBMPの因子のin vivoでの作用にも相違があることが示唆された。
