HGFによる脳神経系の生存維持とHGFからみた神経変性疾患の病態解析

研究概要

増殖因子とそのレセプターが脳において、神経細胞の増殖・分化、脳傷害に対する生存・維持を担うことが知られている。私達はHGFが肝臓をはじめとする各種臓器の再生因子として機能することを明らかにしてきたが、今回、HGFが神経細胞の生存・維持を支える神経栄養因子の一つとして機能することを明らかにした。免疫組織化学ならびにin situハイブリダイゼーションによりHGFならびにMet/HGFレセプターの発現を調べたところ、HGFの発現は前頭葉大脳皮質、扁桃体、海馬、嗅球、小脳、延髄に存在するニューロンや種々の領域のグリア細胞に認められた。一方、Met/HGFレセプターの発現は嗅球、大脳皮質、海馬、一次嗅覚皮質、嗅内野、小脳のニューロンに認められた。また、ラットに実験的脳虚血傷害を引き起こしたところ、HGFならびにMet/HGFレセプターの発現は受傷後12時間後から著しく増加した。さらにHGFはラット胎児脳より分離、初代培養した海馬ニューロンならびにPC-12褐色細胞腫に対しin vitroでの生存を強く促進した。これらの結果はHGFがとりわけ脳傷害に対し、主にパラクリン機構を介して神経細胞の生存・維持を支えるneurotrophic factor(神経栄養因子)の一つとして機能していることを示している。これら結果に基づき、最近、リコンビナントHGFを脳内に投与することにより、脳虚血傷害によって引き起こされる神経細胞死を防ぐことができるかどうか実験動物を用いて解析したところ、HGFの脳内投与によって虚血傷害により神経細胞死が著しく抑制されることを見いだした。
