転写活性化因子PhoBの機能解析

研究概要	大腸菌では培地中のリン酸欠乏によりPhoBに依存して転写誘導のおこる31個以上の遺伝子群がありこれらは総称してリン酸レギュロンと呼ばれている。これら遺伝子群のプロモーター領域には転写活性化因子PhoBのDNA結合の認識配列であるリン酸ボックスが存在している。PhoBとRNAポリメラーゼの蛋白質・蛋白質の相互作用点を明らかにするためリン酸レギュロンの発現を特異的に減少させるRNAポリメラーゼ変異体の分離に成功した。その変異部位はRNAポリメラーゼのσ^<70>サブユニットのhelix-turn-helix上の第一helixにあり、この領域でPhoBとの蛋白質・蛋白質の相互作用に関与している可能性が高いことが示唆された。また、σ^<70>サブユニットの第一helixに部位特異的変異の導入を行い、この部位でのPhoBとの蛋白質・蛋白質の相互作用を確認した。さらに、PhoBとRNAポリメラーゼのリン酸レギュロンプロモーター上での蛋白質・蛋白質の相互作用をさらに明らかにするためにPCR mutagenesis法によりRNAポリメラーゼと相互作用できなくなったと考えられるPhoB変異体の分離を行なった。その部位がPhoB上のある特定領域に集中しており、この領域がRNAポリメラーゼとの相互作用点と考えられた。
