血液細胞の増殖・分化における転写因子E2Fの役割の解析および臨床応用

研究概要	Interferon-α (IFN)感受性のB細胞性白血病細胞株Daudiを用い、IFNによる血液細胞の増殖抑制における転写因子E2Fの役割につき検討した。[結果](1)INF-α 100IU/ml存在下でDaudi細胞株を培養すると、細胞増殖は約24時間後に対照の約20%に抑制された。(2)E2Fのmajor subunitであるE2F-1のmRNA発現は、IFN処理後約8時間より低下した。一方、もう一つのsubunitであるDP-1の発現はほとんど影響を受けなかった。(3)E2F-1のdenovo蛋白合成は、約12時間後より有意に低下した。その結果、E2FのDNA結合活性も減少した。(4)IFN処理24時間後でも、RB遺伝子産物と結合したE2Fはほぼ処理前と変わらずに残存しており、これはIFNによるRBの脱リン酸化に伴うE2F結合の増加によるものと考えられた。脱リン酸化型RBと結合したE2Fは、本来の機能とは逆に転写因子として働くことが知られており、IFN処理後24時間でみられるE2Fは、細胞増殖に関してはnegativeに機能していると推定された。(5)E2F結合によりその活性が調節されるcdc2遺伝子のプロモーターをreporterとして用い、IFNにより実際にin vivoでE2F活性が抑制されるかどうかを調べた。Daudi細胞にelectroporationにてcdc2プロモーターを導入後、IFNを加えて24時間培養し、プロモーター活性の変動をCATアッセイにより測定すると、IFNにより有意に抑制されることがわかった。(6)CMVプロモーターを用いてDaudi細胞にE2F-1を強発現させると、IFNによる増殖抑制効果が減弱した。[結語]以上の結果より、IFNによる血液細胞の増殖抑制にはE2F-1mRNAの早期からの発現抑制によるE2F活性の低下が重要な役割を果たしていることがわかった。この知見は、IFNのより有効な臨床応用、耐性の克服、副作用の防止などに役立つ可能性がある。
