光合成器官の形成と葉緑体機能の発現を制御する核遺伝子に関する研究

研究概要	光合成器官である葉の形成には、葉緑体の正常な分化と発達が必須である。葉緑体の発達と機能の発現を支配する遺伝子の多くは核ゲノムに存在するが、その分子的基盤に関する知見は少ない。本研究では、葉緑体の遺伝情報発現系に関わる蛋白質因子を分離しその機能を調べ、それらの核遺伝子の構造と葉の形成過程における発現様式を明らかにする。今年度、得られた成果は以下の通りである。 1.葉緑体RNA結合蛋白質(RNP)は3つのグループに大別され、それぞれの葉緑体内の役割が異なる可能性を明らかにした。 2.タバコのRNPの大部分はストローマ画分に存在し、30S以下の沈降係数をもつRNA蛋白質複合体として存在する。その複合体には種々のmRNA分子種やrRNA、イントロンをもつpre-tRNAが含まれていることを明らかにした。RNPは葉緑体RNAの安定性に関与していると考えられる。 3.タバコの葉緑体in vitro翻訳系を開発した。光化学系II成分のD1蛋白質をコードするpsbA mRNAがこの系で効率よく翻訳されることを明らかにした。この系を用いれば葉緑体の翻訳過程に関与する制御因子を同定することが可能である。 4.タバコ培養細胞BY2の核抽出液からin vitro転写系を開発した。この反応系でRNAポリメラーゼI、II、IIIによる忠実な転写開始が起こることをプライマー伸長法を用いて確認した。葉緑体関連の核遺伝子の転写因子(光制御因子や器官特異的因子)を分離同定する実験系として活用できる。
