マウス発生工学によるc-mplとトロンボポイエチンを介した巨核球増殖機構の解析

研究概要	マウスの急性白血病ウイルスに由来する癌遺伝子c-mplのリガンドが巨核球の分化増殖と血小板産生を誘導するトロンボポイエチン(TPO)であることが解明された。本研究の目的は、TPOを持続的に産生するトランスジェニックマウスを作製し、そのストローマ細胞を用いて血小板産生を支持する培養系を確立することにある。まず、ニワトリアクチンプロモーターとサイトメガロウイルスエンハンサーを有する発現ベクターpCAGGSにヒトTPOcDNAを連結したDNA(CAG-TPO)を作製した。CAG-TPOの活性は、COS細胞にトランスフェクション後の培養上清の、Ba/F3細胞にc-mpl(TPO receptor)を発現させた細胞に対する増殖活性を測定することにより確認した(東レ基礎研究所玉腰先生、須藤先生との共同実験)。次に、ベクターを除いたDNA断片を切り出し、BDF1(C57BL/6xDBA)マウスの受精卵にマイクロインジェクションした。DNAを注入した480個の受精卵の内生き残り里親に戻したものは403個であり33匹のマウスが4週齢まで育った。それらのtail DNAをPCRで解析したところトランスジーン陽性の個体は得られなかった。トランスジーンが致死的に働いてることを示唆する結果として、1)自然分娩で誕生し翌日に死亡したマウスのうち、DNA解析が可能であった9匹のうち7匹がトランスジーン陽性であった.2)帝王切開したマウスより胎盤を回収しDNA解析をしたところ、7匹中4匹がトランスジーン陽性であった.しかしながらこれらは翌日までに食殺されていた、が得られている。
