温度感受性ヒトp53癌抑制遺伝子を用いた細胞増殖制御機構の解明

研究概要

p53癌抑制遺伝子の変異は殆ど全てのヒト癌で最も高頻度にみられる。我々は野生型p53のもつ細胞増殖機能に解析するために、ヒトp53の138番目をアラニンからバリンに変えた温度感受性変異株(vall38)を作成し、ラット初代線維芽細胞(REF)、ヒト骨肉腫細胞(Saos-2)、ヒトT細胞白血病(Jurkat)、およびヒト赤白血病細胞(K562)に導入した。これらの細胞を低温シフトしたときの細胞増殖に対する影響を、生細胞数、DNA含量等を基に解析したところ、Jurkat細胞はアポトーシスを他の細胞はG1停止をおこしていた。なおSaos-2細胞ではG2/M停止を起こすものもかなりあった。p53によるG1停止に関してはp21cipl(waf1)の発現誘導、cdk/cyclinの阻害、Rbの脱リン酸化、E2Fとの結合を介するDNA合成開始の阻害が考えられている。G1停止をおこす細胞でp53の下流のシグナル伝達をしらべたところ、K562細胞では上記と一致する。REF細胞ではp21cip1(waf1)の発現誘導を介さない、Rbの発現のないSaos-2細胞ではRbの代わりにp130がG1停止の、シグナル伝達に係わっていることが判明した。そこでREF、Saos-2細胞でのG1停止に、またJurkat細胞でのアポトーシスに関与する遺伝子をクローシングするため、低温シフト前と後それぞれ6時間サイクロヘキシミド存在下で培養し、mRNAを精製後cDNAライブラリーを作成した。低温シフト後に野生型活性によって発現誘導されると思われる新たな遺伝子の検索をピオチン/ストレプトアビジンによるサブトラクション法を用いて試みたが得ることが出来なかった。
