研究概要	過剰発現により細胞をがん化するレトロウイルスのがん遺伝子、mafについて解析している。v-Mafやmaf関連遺伝子産物の転写活性化能を解析する実験の過程で様々な細胞にMafあるいはsmall Maf蛋白と別個のbZip蛋白であるp45のヘテロ2量体であるNF-E2などとDNA認識特異性で類似する内在性の極めて強い転写促進活性の存在が観察された。この転写活性化因子は特にNF-E2に類似した認識配列特異性を有するが、細胞の核抽出液をgel shift法で解析したところ、NF-E2やMafのホモ2量体などよりかなり大きな移動度を示した。従ってMafよりも分子量が小さい別個の蛋白がこの活性を担っているものと考えられる。またMaf family蛋白のdominant negative mutantを用いた実験などからも、この内在性の強い転写促進活性の本体はおそらく未知のbZiP蛋白のヘテロ2量体であり、しかもそのcomponentのいずれかはMaf family蛋白と2量体を形成しうる可能性が強く示唆された。そこでこの因子のcloningを試み、現在までにWest-western法、Two-hybrid法により多数のcloneを得て解析中である。既に明らかになっているFos,Junなどとのヘテロ2量体形成に加え、さらに多くのbZip蛋白との相互作用ネットワークを明らかにすることによって、Mafによる細胞がん化の機構が明らかになっていくものと期待される。
