B型肝炎ウイルスX蛋白質と結合する肝細胞内プロテアーゼの機能変化と発癌機序の解析

研究概要

B型肝炎ウイルスによる肝癌発症の引き金となるX蛋白質は、Kunitz型プロテアーゼインヒビターの活性中心構造と類似した構造がその一次構造に見出せることから、何らかの肝細胞内プロテアーゼがX蛋白質と総合作用することが考えられた。以上の考察を発端として我々はこれまでに肝細胞内の細胞質分画にあるプロテアーゼ群から、X蛋白質に結合性を示すプロテアーゼとしてtryptase TL_2 (TL_2)を見出してきた。X蛋白質の中でKunitz型プロテアーゼインヒビターの活性中心構造と類似した部位のアミノ酸偏位体や,欠失体では、TL_2との結合性や、X蛋白質としてのトランス活性化能が失われることが判明している。
本年度の研究で、TL_2がこれまで報告してきたようなプロテアーゼ作用の他に、ATPase作用を持つ多機能蛋白質であることが判明した。なおこのTL_2のATPase作用は、蛋白質の投与により活性化された。最近の研究からX蛋白質は、細胞質から核に移向することが知られており、TL_2のATPase作用が分子シャペロンとしてX蛋白の核内移向に関与する可能性が考えられた。一方TL_2のプロテアーゼ作用が、AMPやGMPで活性化され、ADPやGDPで抑制されることが判明している。これらのことからTL_2のプロテアーゼ作用とATPase作用の解析によるX蛋白のトランス活性化機構と解析している。
