研究概要

1)細胞増殖におけるプロテアーゼ機能の解析:ras遺伝子や増殖因子刺激により合成・分泌が著名に増加するカテプシン前駆体の細胞増殖促進作用と細胞外マトリックス分解作用、カルパインによるnPKC ε down regulationの情報伝達機構における積極的な生理的意義、さらに肝細胞の増殖停止あるいは分化に関与すると考えられる新規膜プロテアーゼの精製と性格づけ、B型肝炎ウイルスX蛋白に結合するトリプターゼ(TL2)のプロテアーゼ機能とX蛋白との相互作用が示された。
2)細胞周期の制御におけるタンパク質分解の役割とその機構の解析:第一減数分裂終了時に限っておこるサイクリンBの緩やかな分解にMos-MAPキナーゼ系とその他未知の因子が関与すること、Mos-MAPキナーゼ系によりcFosがリン酸化され、代謝的安定性が増し、無制限な細胞増殖の誘導につながること、cFosはユビキチン-プロテアソーム系で分解され、このときc-Junの会合と複数の蛋白質キナーゼによるリン酸化が必要であることが明らかになった。
3)細胞外マトリックスの破壊・血管新生におけるプロテアーゼ系の役割の解析:食道癌でのelafinの発現は細胞分裂の亢進した状態から分化の過程に関与している可能性、大腸癌で高頻度に発現している高分子量型の新しいヒトメタプロテアーゼの諸性質、ODC高発現の基底膜浸潤の促進への関与、MMP-9の胃ガンでの血清診断での有効性などが判明した。
