研究概要	アポトーシスは、遺伝子レベルで制御された、核破壊を伴う細胞死の一型で、遺伝子異変などを起こした細胞の増殖を阻止し、それらを殺傷排除することでがんの発生を未然に防ぐ、生体防御の役割を果たしている。このようなことからアポトーシスの機構を解明することは、がんの発生や進展の仕組みを知るだけではなく、がんの進展阻止やがん細胞の殺傷除去に関する方策を考えていく上でも重要である。アポトーシスに関する研究は、このところ急速な進展をとげ、種々のアポトーシス誘導因子や抑制因子が見いだされるとともに、シグナル伝達の仕組みについても分子レベルで検討されるようになってきた。本研究班でも、各班員が独自に開発した実験系を用いて、がん細胞におけるアポートシス発生の機構を多面的に、かつ分子レベルで理解していくことによって、アポトーシスの実態解明に向けた研究が精力的に展開されてきている。例えば哺乳動物細胞には少なくとも一種のイントロンレスmyc遺伝子が存在していることが明らかにされ、これら遺伝子の発現によってグリオーマ細胞に非常に効率よく、しかもc-Mycの場合と異なり野生型p53の発現に依存することなくアポトーシスが誘導されることが見いだされた。またこれらイントロンレスmyc遺伝子をを用いた実験からMyc蛋白質のdeath domainが初めて同定された。またアポトーシス支配因子Fasの生理的なリガンド(FasL)をコードするcDNAがヒトやマウスからも単離され、膜型および細胞質型FasLの生成機構が明らかにされた。さらに酵母の系を用いて活性型可溶性FasLの大量生産に成功し、えられたFasLを用いて種々のモノクローナル抗体が作成され、高感度のELISA法によるFasL検出システムが構築された。そしてこのシステムが、TまたはNK-LGL白血病やNKリンパ節腫瘍の患者血清での可溶性FasL蛋白質の検出に利用できることが示されたことで、がん診断への応用が期待できるようになってきた。この他、Ara-C,Vp-16,CPTなどの抗がん剤によるアポトーシスがTNFや抗Fas抗体によるものと同じくICEファミリープロテアーゼの活性化を伴うことが示されたり、アポトーシスに特異的なDNAの断片化に関与するエンドヌクレアーゼが胸腺細胞から精製され、対応するcDNAがクローニングされるなど、多くの研究成果が挙げられた。
