プロテアソームによる細胞周期の制御

研究概要	細胞周期の制御に関与する様々な因子は代謝的に不安定であり、分解異常による安定性の獲得は無秩序な細胞増殖の要因となる。細胞周期のように多数の因子群の関与した連続的な逐次反応を不可逆的かつ適宜に進行させていくためには、蛋白分解による量的調節が重要である。これらの調節因子群の細胞内動態を監視して細胞周期の進行を統御している細胞内蛋白質分解システムはユビキチンとプロテアソームから構成されている。我々は数年来、分子量約200万の巨大なATP依存性プロテアーゼ複合体であるプロテアソームの構造と機能に関する研究を続けており、本年度、細胞周期制御におけるプロテアソームの役割を解明する研究を行い以下の研究成果を得た。酵母プロテアソームを構成する調節ユニットの遺伝学的な機能解析から、本酵素がG1/S転移、M期への進入、M期からの脱出など細胞周期の複数の段階で機能していることを明らかにした。この結果、蛋白分解がG1/S転移、M期の進行に必須であり、代謝的安定性の変動による細胞周期因子の再編成が細胞周期制御において重要であるが示唆された。本年度はさらに、プロテアソームによるプロトオンコジーンcFosやODC(オルニチン脱炭酸酵素)の分解調節機構を明らかにした。その結果、これらの転写因子や代謝調節酵素の細胞内レベルはプロテアソームによって厳格に統御されており、その代謝的安定性の破綻が、無制限な細胞増殖の誘導や細胞の腫瘍化の原因となる可能性が示唆された。その他、プロテアソームの特異的な阻害剤ラクタシスチンを用い、本酵素がPDGF(血漿板由来細胞増殖因子)レセプターのユビキチン経路による分解を介した脱感作機構や細胞死の誘導調節に関与していることも証明した。さらに、遺伝子構造解析研究からプロテアソームが内在性抗原のプロセッシング酵素として細胞性免疫の発動に密接に関係していることを示唆し、この免疫調節機能を担う酵素を免疫プロテアソームと命名した。この結果、免疫プロテアソームが免疫始動制御のキ-酵素であることが判明した。
