癌細胞の自律性増殖におけるTGF-βのシグナル伝達機構の役割

研究概要

1.TGF-βレセプターの腫瘍細胞での発現と癌化 : TGF-βのII型レセプターは遺伝性大腸癌などで突然変異が起こることから、重要な癌抑制遺伝子と考えられている。我々は胃癌の組織でのTGF-βレセプターの発現を検討し、胃癌組織ではむしろTGF-βレセプターは増えている所見を得た。一方、前立腺癌の細胞株LNCaPではTGF-βのI型レセプターの発現が無くなっていることを確認した。
2.TGF-βIII型レセプターのリガンドとの結合部位 : TGF-βIII型レセプターのリガンドとの結合部位をIII型レセプターの変異クローンを用いて検討し、結合部位にはN末端近傍とC末端近傍の2カ所があることを確認した。
3.TGF-βレセプターの活性化のメカニズム : TGF-βのI型レセプター活性化にはGSドメインとよばれる特異的構造が重要であることが知られている。今回我々は、I型レセプターの膜貫通領域とGSドメインの間に位置する領域 (JM領域) に位置する172番目のセリンや176番目のスレオニン残基を置換すると細胞の増殖抑制シグナルは伝達されなくなるのに対し、細胞外マトリックスの産生は正レセプターと同様に伝えられることを確認した。
4.TGF-βI型レセプターの細胞内基質の同定とシグナル伝達機構 : 我々は酵母を用いたtwo-hybrid法によってTGF-βI型レセプターの基質のクローニングを試み、farnesyl transferase-α (FT-α) がTGF-βのI型レセプターと結合することを見いだした。FT-αはRasなどの低分子量G蛋白質に作用し、膜との結合を促進する酵素である。FT-αは活性化されたI型レセプターとより強く結合し、またI型レセプターによってリン酸化されることが明らかになった。
