腫瘍でのADF/チオレドキシンの活性化機構並びに抗癌剤の影響と作用機序の解析

研究概要	HTLV-I感染細胞株培養上清中より分離精製されたATL由来因子(ADF)は大腸菌の還元補酵素チオレドキシン(TRX)のヒト相同体であり、その活性部位にあるシスチン残基を介した酸化還元反応により細胞内外の酸化還元環境の維持(レドックス制御)を行っている。ADF/TRXはその還元活性により多彩な機能を有する因子として報告されているが、本研究では、ADF/TRXが多くの腫瘍組織において高発現している事実に着目し、細胞の腫瘍化とADF/TRXはじめとする細胞内レドックス制御機構との関連を解析するとともに、腫瘍細胞の抗癌剤耐性獲得におけるADF/TRXの関与について検討した。さらには、悪性腫瘍患者の血清中ADF/TRXの定量法を開発し、診断、予後判定などの臨床医学における有用性について検討した。その結果、1、シスプラチン、アドリアマイシンなどの活性酸素を産生する抗癌剤に対する耐性と細胞内ADF/TRXの発現量との間に相関を認めた。2、シスプラチン、ブレオマイシンなどの抗癌剤がADF/TRXを発現誘導することを見い出した。また、ADF/TRXを遺伝子導入した細胞株はシスプラチンに対する耐性を獲得したが、これにはADF/TRXの活性酸素消去作用が関与しているものと示唆された。3、抗ADF/TRX抗体を用いたELISAサンドウイッチ法による血清ADF/TRXの定量法を開発し、肝細胞癌患者の血清中においてADF/TRXが有意に高値を示すことを見い出した。4、活性酸素消去系酵素の発現が、癌細胞の増殖能増強、浸潤・転移能の獲得などの生物学的悪性化の進展に関与していることを見い出した。
