IP_3誘導Ca^<2+>放出チャネルの構造・機能相関

研究概要

IP_3受容体はIP_3によって活性化される細胞内Ca^<2+>放出チャネルで、その構造・機能を解析した。
1.タイプ1のIP_3結合領域(アミノ末端側734アミノ酸)を大腸菌で発現させる系を開発した。IP_3は負のリン酸基を3つもつので、この領域内にある正のLysとArgを部位特異的置換することによりリガンド結合アミノ酸を解析した。その結果、Arg-265、Lys-508、Arg-511はIP_3結合に必須であった。また、Arg-568のGlnへの置換はIP_3と1,3,4,5-IP_4の結合親和性を低下させた。
2.カルモジュリン結合領域を解析した。タイプ1では、cAMP依存性キナーゼのリン酸化基質となるSer-1588の近傍の配列(Lys-1564からArg-1585)が結合に必須であった。タイプ2の類似した配列(Lys-1565からArg-1587)も結合した。タイプ3ではこの配列が存在せず、カルモジュリンによる制御の違いが示唆された。
3.IP_3誘導Ca^<2+>放出(IICR)のキネティクスを精製受容体/人工リポソーム/fluo-3系で解析した。IICRは二相性(速い相(f相)と遅い相(s相))を示し、初速度のEC_<50>は100nM IP_3であった。Hill係数は1.8と正の協同性を示した。f相の見かけの速度定数(k_<fast>=0.3-0.7s^<-1>)はs相(k_<slow>=0.03-0.07s^<-1>)の約10倍であった。f相によるCa^<2+>放出はIP_3濃度に依存して増加(量子的放出)を示したが、s相によるCa^<2+>放出量はほぼ一定であった。これより、IP_3受容体は2つのIP_3結合状態をもち、f相は低親和性、s相は高親和性の状態であることが示唆された。
4.カビの代謝産物であるアデノフォスチン(AP)は、拮抗的でより強力なアゴニスト作用をもつ。AP誘導Ca^<2+>放出もIICRとよく似たキネティクスを示したが、高い協同性(Hill係数3.9)があった。〔^3H〕-IP_3結合の拮抗阻害実験では、IP_3に協同性はないが(Hill係数1.1、Ki=41nM)APは正の協同性(Hill係数1.9、Ki=10nM)を示した。
