グルタミン酸受容体を発現した細胞株における細胞内情報伝達の分子機構

研究概要	細胞内外のCa^<2+>と細胞死の制御機構の関係につき検討した。グルタミン酸やそのアナログは神経細胞では細胞内Ca^<2+>濃度を上昇させることにより細胞死を引き起こすことが知られている。我々はイオン作動性グルタミン酸受容体を機能的に発現しテイルMIN6細胞ではグルタミン酸やそのアナログであるNMDA処置により、細胞内Ca^<2+>が上昇しても細胞死が誘導されないことを示した。一方、細胞外のCa^<2+>をEGTAやBAPTAでキレートするとBax mRNAの発現が増強すると同時に細胞のアポトーシスが誘導された。この際、共焦点レーザー顕微鏡を用いた画像解析により、細胞質内および核内のCa^<2+>が減少することが示唆された。また、Bax cDNAをプローブとして実施したノザン法により、これまで報告のない約4.5kbの転写物を認めた。細胞外のCa^<2+>をキレートしE場合、この4.5kbの転写物の発現が増強した。これらの事実より、Bax遺伝子の発現は細胞内Ca^<2+>の濃度により調節されている可能性が示唆された。次にCa^<2+>によるBax遺伝子の転写調節の制御を検討する目的で、我々はマウスのゲノムlibraryをスクリーニングし、Bax遺伝子のプロモーター領域を含む4.2kbのDNA断片を単離した。現在、転写開始点を決定し、CATアッセイ法を用い、Ca^<2+>によるプロモーター活性の制御機構について検討している。同時に4.5kbの転写物のクローニングを試みている。
