内向き整流特性カリウムチャネルの神経伝達調節と可塑性における役割

研究概要

中枢神経系における内向き整流性カリウムチャネルの分子機構を明らかにするために、マウス脳よりまったくあたらしい2回膜貫通型内向き整流性カリウムチャネルcDNA(K_<AB>-2,the second type of inward rectifying K^+ channel with an ATP-binding domain)をクローニングした。アフリカツメガエル卵母細胞発現系でK_<AB>-2チャネルは、古典的な内向き整流特性を示すカリウム電流を示した。in situ hybridization法による解析の結果、K_<AB>-2mRNAは脳幹部神経核とともに、脳梁や小脳白質などのグリア細胞に強く発現していた。さらに、網膜のミューラー細胞やアストロサイト初代培養にもK_<AB>-2チャネルが発現しており、グリア細胞におけるK^+-buffering systemに関与していると考えられる。また、免疫組織化学の結果、K_<AB>-2チャネルは腎臓遠位尿細管細胞や内耳血管条基底細胞の基底外側に発現していた。以上のことからK_<AB>-2チャネルは神経系グリア細胞や腎臓、内耳において細胞内外のK^+イオンの輸送に関与していると示唆される。現在、特にグリア細胞K^+-buffering systemにおけるK_<AB>-2チャネルの生理的役割を明らかにするために、ES細胞を用いた発生工学的手法によりK_<AB>-2チャネルのノックアウトマウスの作成を試みている。
