研究概要

モルテングロビュール(MG)は球状タンパウ質に共通する折り畳み中間体であると考えられている。この状態は構造的には厳密に定義された状態でありながら、現実の蛋白質のMGではそのサイズ、形状あるいは二次構造のパッキングの程度(tertiary fold)など定量的な評価があまりなされていなかった。そこで、典型的なMGとしてとらえられているアポミオグロビン、チトクロムc及びαラクトアルブミンのMG状態について、X線溶液散乱によりその構造の特徴を調べた。MGの慣性半径は天然状態の慣性半径から25〜30%大きくなっていたが、αラクトアルブミンではその増加の程度は11〜15%であった。Ptitsynらは慣性半径の増加は10%以下であると主張していたが、慣性半径の増加は30%までにも及ぶことが明らかになった。αラクトアルブミンでは分子内SS結合によって増加の程度が押さえられていると推測される。
MGは距離分布関数(P(r))によって二種類に分類されることが示された。一つはチトクロムcやαラクトアルブミンのMGのように一相性のP(r)を示すものであり、他はアポミオグロビンのMG等のように二相性のP(r)を示すものである。前者ではかなりの程度tertiary foldが完成しているが、後者ではごく一部ができているにすぎない。tertiary foldは、チトクロムcでは疎水相互作用が、αラクトアルブミンではSS結合によって保持されていると推測される。また、二相性のP(r)を示す構造は、MGのみならず折り畳みの初期に形成される構造に特徴的なものであると考えられる。
天然構造とコンパクトな変性状態での蛋白質の動的性質を理解するために、中性子非弾性散乱測定を行い、非弾性散乱スペクトルに差を検出した。詳細な解析は今後の課題である。
