研究概要	減数分裂の開始と進行の制御機構につき次のことを明らかにた。(1)分裂酵母の減数第一分裂を誘導するためには制御因子Mei2(RNA結合蛋白質)とmeiRNAと呼ぶRNA分子の協同作業が必要である。meiRNAの果している機能を探るため,meiRNAを欠く細胞に高発現させると減数分裂を誘導できる分裂酵母遺伝子を検索した結果、減数分裂時のみに特異的に発現する微小管結合蛋白質をコードする遺伝子ssm4を同定た。Ssm4蛋白質はN末端に微小管結合領域、C末端にコイルドコイル構造をもち、栄養生長には必要とされないが、Ssm4を欠く細胞は減数分裂過程に異常を示し、しばしば2個しか胞子を含まない胞子嚢を生じた。(2)アフリカツメガエル卵から開発した減数分裂周期を再現する無細胞系(meiotic extracts)を用いて、減数分裂間期においてMos蛋白質がS期の抑制をもたらす機構を解析したところ、MAPKKK活性を持つMosの下流ではMAPキナーゼを介する経路と介さない経路とが機能しており、前者の経路はcyclin Bの蛋白質合成に関わっている可能性が明らかとなった。この観察は昨年度報告したin vivo実験の結果を補強するものである。(3)アフリカツメガエルの卵成熟機構を再検討した結果、従来信じられていた三つの説に誤りを認めた。第一に、卵成熟の開始にcAMP依存性プロテインキナーゼ(PKA)活性の低下は必須条件ではなかった。第二に、Mosが分裂停止因子(CSF)活性を発揮する際にcdk2キナーゼ活性は関与していなかった。第三に、多年にわたり信じられてきた、卵成熟に核の内容は関与しないという説に対して反証が得られた。
