研究概要

化学療法は癌の有効な治療法のひとつであるが、ほとんどの抗癌剤が効かなくなる多剤耐性の問題は癌化学療法の大きな障害となっている。一方、耐性黄色ブドウ球菌(MRSA)に代表されるように、感染症の治療に有効な抗菌剤に対して耐性を獲得した病原菌が出現し、大きな社会問題となっている。本研究では、植物、微生物、海洋生物など広範囲の天然物を素材として、MRSAをはじめとする耐性菌に有効な抗菌物質の探索、多剤耐性癌に有効な耐性克服作用をもつ化合物の探索、薬剤耐性克服作用をもつ天然物の化学合成や化学修飾、薬剤耐性発現機構の解明とそれに基づく耐性克服物質の開発を検討した。以下に本年度の研究成果の概要を報告する。
1)薬用植物、海洋動物、微生物の抽出物について、スクリーニングを行った結果、日本産イチイより数種のP-糖蛋白質機能阻害物質を単離し、これらの化合物がアジトピンのP-糖蛋白質への結合を競合的に阻害することを見い出した。2)環状ペプチドRAの全合成ならびに環状ペプチド・アスチン類の化学修飾を行い、抗腫瘍活性の評価を行った。3)ウスチロキシンDの化学変換による活性構造の解析、キュラシンAの部分構造の合成による絶対構造の決定と活性評価、アレナスタチン類のチューブリンへの作用、新規タキソイドのチューブリンへの活性評価を行った。4)薬剤耐性癌細胞に効果を示す物質を微生物代謝産物より単離した。5)耐性の治療においては、新しいキノリン化合物MS-209やシクロスポリン誘導体PSC833、チアパミール誘導体Roll-596D、等が耐性克服に有効であることが見い出された。6)タキサン化合物の13位のエステル部分の種々の光学活性誘導体の一般合成法を開発した。7)抗結核剤であるリファンピシンに対する抗酸菌の耐性機構として、リボシル化という新しい不活性化機構を見い出し、この機構が多くの非結核性の抗酸菌に存在することを証明した。8)ビフェニルエーテルの新しい構築法を開発し、セコアグルコバンコマイシンの合成に成功した。また、合成セコアグルコバンコマイシンが細胞壁モデルのN-Ac-D-Ala-D-Alaを分子認識することを明らかにした。
