天然物を素材とした薬剤耐性克服薬の探索と創製

研究概要	化学療法は癌の有効な治療法のひとつであるが、ほとんどの抗癌剤が効かなくなる多剤耐性の問題は癌化学療法の大きな障害となっている。一方、耐性黄色ブドウ球菌(MRSA)に代表されるように、感染症の治療に有効な抗菌剤に対して耐性を獲得した病原菌が出現し、大きな社会問題となっている。本研究では、植物、微生物、海洋生物など広範囲の天然物を素材として、MRSAをはじめとする耐性菌に有効な抗菌物質の探索、多剤耐性癌に有効な耐性克服作用をもつ化合物の探索、薬剤耐性克服作用をもつ天然物の化学合成や化学修飾、薬剤耐性発現機構の解明とそれに基づく耐性克服物質の開発を検討した。以下に本年度の研究成果の概要を報告する。 1)日本産イチイおよび中国産イチイより数種の新規タキサン化合物を単離し、それらのうちのいくつかにP-糖蛋白質機能阻害作用を見い出し、in vivoでの耐性克服作用も認められた。2)環状ヘキサペプチドRAの構造活性相関の検討により、抗腫瘍性環状ペプチドRA-Vllを開発し、臨床第1相試験を行った。3)キュラシンAの全合成とその類縁体の構造活性相関、ウスチロキシンDの化学変換による活性構造の解析を検討した。4)薬剤耐性癌細胞に効果を示す物質を放線菌ならびに植物より単離した。5)キノリン誘導体MS-209が、in vivoで多剤耐性白血病細胞に対する抗がん剤の治療効果を増強することを明らかにした。また、PSC833の多剤耐性克服能が抗P-糖タンパク抗体MRK-16により増強されることを見い出した。6)抗結核剤であるリファンピシンに対する抗酸菌の耐性機構として新たに見い出したリボシル化が、抗酸菌のみならず多くの病原性放線菌においても重要な役割を果たしていることが明らかとなった。リボシル化に関与する遺伝子がクローニングされ、さらにリボシル化の中間体と考えられるADP-リボシル化したリファンピシンの存在が明らかとなった。7)独自に開発したTTN酸化法を用いて、セコアグルコバンコマイシン類を合成し、本化合物がバンコマイシン耐性菌に存在する細胞壁モデルのペプチド鎖と強い分子間水素結合を形成することを明らかにした。多剤耐性克服活性をもつハパロシンの全合成を行った。
