研究概要

昨年度に引き続き、1)細胞外基質分子とインテグリンの間の分子認識機構、2)インテグリンの下流のシグナル伝達分子の同定およびその機能、3)インテグリンを介する細胞の増殖・分化の制御機構、の3点に焦点を絞って研究を進め、以下の研究成果を得た。
【1】インテグリンα5β1を介する細胞接着がリガンド分子(フィブロネクチン)の選択的スプライシングにより制御されていることを発見した。(関口)【2】インテグリンα3β1とその特異的リガンド(ラミニン-5)の結合には、リガンド分子上のG2ドメインが関与することを組換えフラグメントおよび合成ペプチドを用いて同定した。(宮崎)【3】インテグリンからのシグナルを細胞骨格系に伝達するパキシリンには、選択的スプライシングによるアイソフォームが3種類あること、その一つのγタイプはビンキュリンとの結合活性がないことを明らかにした。(佐邊)【4】インテグリンα3β1と複合体を形成している膜結合型細胞増殖因子HB-EGFのヘパリン結合領域がその活性発現に重要な働きをしていることを明らかにした。(目加田)【5】平滑筋細胞の分化形質の維持にラミニンと増殖因子IGF-1の両方が必要であることを明らかにするともに、インテグリンα1β1が平滑筋細胞とその関連組織に選択的に発現することを発見した。(祖父江)【6】細胞-細胞間接着分子カドヘリンとインテグリンとが相互に機能的に連絡していることをカドヘリン遺伝子導入細胞を用いて示すことに成功した。また、カドヘリンを血球系細胞に強制発現させると、細胞増殖の抑制がおこることを見いだした。(小澤)
