研究概要

SecGはSecAと機能的に相互作用し、SecAの膜内への挿入・脱離に共役して膜内配向性の反転・復帰を繰り返すことを明らかにした(徳田)。
SecY-SecE-SecG複合体の相互作用を、ヒスチジンタグ法を用いて明らかにした。SecY-SecA間の相互作用部位を変異体を用いて明らかにした。FtsHがSecYのクオリティーコントロールに果たす役割を明らかにした(伊藤)。
葉緑体ゲノムにコードされたシトクロムfのチラコイド内への輸送にはSecAが関与していることを明らかにした。エンドウ葉緑体SecYがチラコイドの内在性膜蛋白質であることを証明した(遠藤)。
リン脂質平面膜にSecY,SecEを再構成し、SecY/Eより構成されたイオンチャンネルの検出に成功した。チャンネルのコンダクタンスは約100pSであり、SecAとATP、前駆体蛋白質の添加で順次閉鎖することを見出した(平田)。
不連続蛋白質膜透過反応は、前駆体蛋白質の疎水性領域に依存するものであることを明らかにし、SecYによって疎水性領域が認識されることを示した(森)。
枯草菌シグナル認識粒子の構造と機能を明らかにした。さらにSRP複合体と前駆体蛋白質や蛋白質合成身長因子EF-Gとの相互作用を明らかにした(山根)。
大腸菌の蛋白質膜透過装置を新たな視点から再構成することを試み、極めて活性の高い再構成プロテオリポソームを得た。再構成活性は、SecA,ATP,SecB,シグナルシークエンスに依存であることを明らかにした(渡邊)。
